What DVD ripping software you've used?

When you made the backup using DVD Shrink, did you re-author your DVD or uncheck some of the Audio boxes? This happened to me once while trying to get the compression ratio as low as I could, I unchecked ALL the Audio boxes and well...no audio. :)

It could be that the dvd in question is by default DTS audio. Then you need to toggle through your available audio streams to find the one that plays, using your remote.
